第一夫人办公室（英语：Office of the First Lady，缩写：OFL）是对美国第一夫人负责的白宫幕僚。办公室本身并无任何宪制职责，但随着第一夫人在美国历史上的角色增进而变得越发重要[2]。第一夫人办公室是美国总统行政办公室辖下白宫办公厅的一个组成部分[3]，其办公地点座落于白宫东厢办公室（英语：East Wing）。

第一夫人的角色在美国历史上一直都相当重要，但直至1900年代前白宫都没有专属于第一夫人的幕僚团队。贝尔·哈格纳是首位属于第一夫人的幕僚，在1902年10月2日被伊蒂丝·罗斯福聘请为白宫社交秘书（英语：White House Social Secretary）[4]。爱莲娜·罗斯福首次将其幕僚团队扩展至社交及行政秘书以外的职责，并任命马尔维娜·汤普森（英语：Malvina Thompson）为其私人秘书[5]；杰奎琳·肯尼迪则是首位聘用新闻秘书的第一夫人[6][2]。在罗莎琳·卡特在任期间，第一夫人的幕僚团队正式冠以“第一夫人办公室”的名称，并第一次设幕僚长一职及迁至白宫东厢办公室（英语：East Wing）办公[1]。
