蘭開斯特里亞號於1940年6月17日鄧寇克行動後，乘載然留在法國的六千至九千名的工作人員、軍人和平民，在法國聖納澤爾港外海被納粹德國空軍的轟炸機擊沉。據推測這次沉沒事件導致的罹難人數達4000以上，只有二千五百人獲救。這次事件是英國航海史上死亡最多的單次事件，也是英國在二戰中傷亡最慘重的一次事件。據稱蘭開斯特里亞號的罹難人數比鐵達尼號（Titanic）和露西塔尼亞號（Lusitania）罹難人數的總數還多。
