维甲酸诱导基因-I[5](RIG-I,retinoic acid-inducible gene I)是脊椎动物细胞质中的一种模式识别受体(PRR),结构上属于DExD/H-box类RNA解旋酶[6][7][8],在人类基因组中由DDX58基因编码[9],其N端有两个CARD结构域,C端则可与RNA结合,为先天免疫反应中的重要蛋白[6][7]。
RIG-I平时处于灭活状态,在细胞被西尼罗河病毒、黄热病毒、日本脑炎病毒、甲型流感病毒、仙台病毒与冠状病毒等RNA病毒感染后[6][10],RIG-I可与病毒的双股RNA(dsRNA)结合而被激活,接着即可以CARD结构域和线粒体外膜上的线粒体抗病毒信号蛋白(MAVS)结合[7],启动第一型干扰素(IFN I)途径的免疫反应[6],后者可阻止病毒扩散、造成发炎反应以及协助激活后天免疫[11]。除识别病毒RNA外,在不同癌细胞中RIG-I还能启动其他反应,包括促进或抑制肿瘤增殖(因肿瘤类型而异)、激活B细胞、影响细胞分化等[12]。有些无脊椎动物(牡蛎)也具有和RIG-I与MAVS同源的蛋白,且可能也有抗病毒的免疫功能[13]
